Diabrotica speciosa (Ger.) (Coleoptera: Chrysomelidae): New Pest in Table Grape Orchards Neotropical Entomology
ROBERTO,SÉRGIO R.; GENTA,WERNER; VENTURA,MAURÍCIO U..
We report for the first time the occurrence of Diabrotica speciosa (Ger.) damaging blooming clusters in grapevine orchards in Northwest Paraná State. On middle September, 2000, insects were found feeding on style, stigma and eventually on ovary. Up to 15 adults per cluster were observed. Insects feeding on stigma caused flowers aborting. In high population areas, clusters showed low number of fruits and were out of market standards.

Expression of Grapevine leafroll-associated virus 3 coat protein gene in Escherichia coli and production of polyclonal antibodies Trop. Plant Pathol.
Fajardo,Thor V.M.; Barros,Danielle R.; Nickel,Osmar; Kuhn,Gilmar B.; Zerbini,F. Murilo.
Grapevine leafroll-associated virus 3 (GLRaV-3), the main viral species of the grapevine leafroll complex, causes yield and quality reduction in grapes (Vitis spp.). The coat protein gene was RT-PCR-amplified from total RNA extracted from infected grapevine leaves and the amplified fragment was cloned and completely sequenced. The fragment was subsequently subcloned into the pRSET-C expression vector. The recombinant plasmid was used to transform Escherichia coli BL21:DE3 and express the capsid protein. The coat protein, fused to a 6 His-tag, was purified by affinity chromatography using an Ni-NTA resin. The identity of the purified protein was confirmed by SDS-PAGE and Western blot. The in vitro-expressed protein was quantified and used for rabbit...

Garlic extract improves budbreak of the 'Niagara Rosada' grapevines on sub-tropical regions Ciência Rural
Botelho,Renato Vasconcelos; Pires,Erasmo José Paioli; Moura,Mara Fernandes; Terra,Maurilo Monteiro; Tecchio,Marco Antonio.
In the regions with mild winters, usually the chilling requirements for bud dormancy release of grapevines are not fulfilled, and only the highly toxic products CaCN2 (calcium cyanamide) and H2CN2 (hydrogen cyanamide) are recommended in conventional viticulture for budbreak. The purpose of this study was to verify the effect of garlic extract on budbreak of 'Niagara Rosada' grapevines in two different vineyards. The highest dose of garlic extract (70mL L-1) showed a great potential for budbreak in organic production, presenting similar effects of cyanamides in budbreak. This treatment improved the sprouting percentage, number of clusters, accelerated the beginning of sprouting and reduced the cycle between pruning and harvest.

Genetic changes in grapevine genomes after stress induced by in vitro cultivation, thermotherapy and virus infection, as revealed by AFLP Genet. Mol. Biol.
Baránek,Miroslav; Raddová,Jana; Krizan,Bretislav; Pidra,Miroslav.
The Amplification Fragment Length Polymorphism (AFLP) technique was employed to study genetic variations which can be induced in vines by the stress occurring during different aspects of viticulture (in vitro cultivation, in vitro thermotherapy and virus infection). Analysis of AFLP banding patterns, generated by using 15 primer combinations, pointed to negligible genetic variation among plants exposed to individual stress. The average of similarity coefficients between differently stressed plants of the cultivars Müller Thurgau and Riesling were 0.984 and 0.991, respectively, as revealed by AFLP analysis. The low incidence of observed polymorphism demonstrates the high level of genome uniformity in plants reproduced by in vitro micropropagation via nodes,...

Genetic diversity in table grapes based on RAPD and microsatellite markers PAB
Leão,Patrícia Coelho de Souza; Motoike,Sérgio Yoshimitsu.
The objective of this work was to analyze the genetic diversity of 47 table grape accessions, from the grapevine germplasm bank of Embrapa Semiárido, using 20 RAPD and seven microsatellite markers. Genetic distances between pairs of accessions were obtained based on Jaccard's similarity index for RAPD data and on the arithmetic complement of the weighted index for microsatellite data. The groups were formed according to the Tocher's cluster analysis and to the unweighted pair‑group method with arithmetic mean (UPGMA). The microsatellite markers were more efficient than the RAPD ones in the identification of genetic relationships. Information on the genetic distance, based on molecular characteristics and coupled with the cultivar agronomic performance,...

Genetic diversity of a brazilian wine grape germplasm collection based on morphoagronomic traits Rev. Bras. Frutic.
Leão,Patrícia Coelho de S.; Cruz,Cosme Damião; Motoike,Sérgio Y..
The objectives of this study were to evaluate the performance of cultivars, to quantify the variability and to estimate the genetic distances of 66 wine grape accessions in the Grape Germplasm Bank of the EMBRAPA Semi-Arid, in Juazeiro, BA, Brazil, through the characterization of discrete and continuous phenotypic variables. Multivariate statistics, such as, principal components, Tocher's optimization procedure, and the graphic of the distance, were efficient in grouping more similar genotypes, according to their phenotypic characteristics. There was no agreement in the formation of groups between continuous and discrete morpho-agronomic traits, when Tocher's optimization procedure was used. Discrete variables allowed the separation of Vitis vinifera and...

Phosphorus fractions in soil cultivated with vineyards after 62 years of poultry litter addition PAB
Schmitt,Djalma Eugênio; Gatiboni,Luciano Colpo; Heidemann,Joice Crescêncio; Dall’Orsoletta,Daniel João; Boitt,Gustavo; Brunetto,Gustavo.
Abstract: The objective of this work was to evaluate changes in phosphorus fractions in a Humic Cambisol cultivated with vineyards, after 62 years of additional fertilization with poultry litter. Soil samples were collected from two vineyards (V1 and V2) and a forested area adjacent to them. A total of 1,365 and 910 kg ha-1 P were applied to V1 and V2, respectively, throughout the past 62 years. Samples were collected at 0-10, 10-20, 20-30, and 30-40-cm soil depths for the analysis of soil physicochemical properties. The addition of poultry litter to the soil of the vineyards increased the contents of inorganic P (Pi) in all fractions, except in the residual fraction (residual-P), mainly at the 0-10-cm soil depth, in comparison with the forested area. The...

Repeatability coefficients and genetic gains in table grape progenies for the Brazilian semi-arid region Scientia Agricola
Leão,Patrícia Coelho de Souza; Nunes,Bruna Thaís Gonçalves; Souza,Emille Mayara Carvalho de.
ABSTRACT: Table grape stands out among the main fruit-bearing plants of irrigated agriculture in the São Francisco Valley region of Brazil. This study estimated repeatability and heritability coefficients and predicted genetic gains in order to select superior genotypes in grape progenies from controlled hybridizations using the Restricted Maximum Likelihood/Best Linear Unbiased Prediction (REML/BLUP) methodology. Individual plants were evaluated for the variables of production (kg per plant), number of bunches, bunch weight (g), berry diameter (mm), and soluble solids content (°Brix). We evaluated 194 hybrids from 30 crosses between Vitis vinifera and interspecific hybrids in Juazeiro, Bahia State, Brazil, over four growing seasons. Repeatability...

Root distribution of irrigated grapevine rootstocks in a coarse texture soil of the São Francisco Valley, Brazil Rev. Bras. Frutic.
BASSOI,LUÍS HENRIQUE; GRANGEIRO,LEILSON COSTA; SILVA,JOSÉ ANTONIO MOURA E; SILVA,EMANUEL ELDER GOMES DA.
An experiment was carried out to determine the root distribution of four grapevine rootstocks (Salt Creek, Dogridge, Courdec 1613, IAC 572) in a coarse texture soil of a commercial growing area in Petrolina County, São Francisco Valley, Brazil. Rootstocks were grafted to a seedless table grape cv. Festival, and irrigated by microsprinkler. Roots were quantified by the trench wall method aided by digital image analysis. Results indicated that roots reached 1 m depth, but few differences among rootstocks were found. All of them presented at least 90 % of the roots distributed until 0.6 m depth, with a greater root presence in the first 0.4 m. The upper 0.6 m can be taken into account as the effective rooting depth for soil and water management.
